چکیده

The aim of this study was to evaluate in vitro the cytotoxicity on human periodontal ligament fibroblasts of three root-end filling materials: MTA Angelus®, EndoSequence Root Repair Material Putty® and Super EBA®. A primary culture of human periodontal ligament fibroblasts was previously obtained in order to evaluate the cytotoxicity of the three extracts from the root-end filling materials after 2 and 7 days of setting. Serial dilutions of these extracts (1:1, 1:2, 1:4 and 1:8) were evaluated at 1, 3 and 7 days using the methyl-thiazol-tetrazolium (MTT) colorimetric assay. Cell viability was evaluated as percentage of the negative control group, which represented 100% cell viability. Statistical analyses were done with t-test, ANOVA and Kruskal-Wallis test at a significance level of 5%. It was found that the main difference among root-end filling materials was in the higher dilutions (p<0.05), but there was a similar behavior in lower dilutions (p>0.05). Cell viability of MTA Angelus® was superior for 2-day setting (p<0.05), compared with the other two root-end fillings. There were no statistically significant differences between 7-day set MTA Angelus® and EndoSequence Root Repair Material Putty®. Super EBA® showed the lowest percentage of cell viability at higher dilutions (p<0.05). Therefore, MTA Angelus® and EndoSequence Root Repair Material Putty® were less cytotoxic in the highest dilution (1:1) compared with Super EBA®.

Attachment of human periodontal ligament fibroblasts to 3 different root-end filling materials: Scanning electron microscope observation.

OBJECTIVE The attachment behavior of the human periodontal ligament (PDL) fibroblasts to root-end filling materials (amalgam and Super-EBA) was compared in vitro to gutta-percha by means of scanning electron microscope. STUDY DESIGN Amalgam and Super-EBA were placed in a prepared cavity of root slices of freshly extracted human teeth and evaluated freshly prepared. Root slices of teeth with c...

بررسی میزان بیان (expression فیبرونکتین، کلاژن نوع I و   TGF-ß)وسط سلول‌های فیبروبلاست لیگامان پریودونتال انسانی در مجاورت مواد پر کننده انتهای ریشه

Background and Aim: Several materials have been introduced for retrograde fillings, pulp capping and sealing root perforations, but their biological effect on vital tissues and cells is not clear. The purpose of this study was to evaluate the reaction of human periodontal ligament fibroblasts to four root canal filling materials: Pro Root MTA, Root MTA, Portland cement and amalgam. Materials a...

Cytotoxicity of mineral trioxide aggregate using human periodontal ligament fibroblasts.

The purpose of the present study was to compare the cytotoxicity of mineral trioxide aggregate (MTA) to other commonly used retrofilling materials, Super-EBA and amalgam. This was accomplished using a cell viability assay for mitochondrial dehydrogenase activity in human periodontal ligament fibroblasts after 24-hr exposure to extracts of varying concentrations of the test materials, in both fr...
